What happens if you settle on a mine or rare herb location?

If you have miners or herbalists, do they automatically harvest?

I think that the resources would just disappear. As for automatic harvesting, that would be a definite "No" based on the answer I got from the Devs on a similar question about automatic harvesting and sovereignty.

The resources will disappear and appear some other "random" place. Much like if you deplete a herb spot.

What would happen if there is a harvester or a caravan harvesting on a spot, and a settler or an exodus is coming to that very square? Will the resource gatherers be bumped away?

Yes, the harvester would be bumped, although many spots where resources appear are not settleable anyway.  (Such as resource spawn spots.)
